Enrico Manfrini studied at the Academy of Bologna, where he met and formed a friendship with the sculptor Cesarino Vincenzi who was also a student at the Academy, both of whom had a positive mutual artistic influence, subsequently Manfrini moved to the Academy of Fine Arts of Brera at the chair of the sculptor Francesco Messina, of whom, upon obtaining his diploma, he would become assistant for a quarter of a century, finally succeeding his master: he would retain the chair until 1984.

He was a member of the Accademia Clementina in Bologna and the Accademia di S. Luca in Rome. He was part of the Superior Council of the Ministry of Fine Arts under the Spadolini government. In 1983 he was awarded the title of Commander of the Italian Republic. He was also appointed Knight Grand Cross of the Order of Saint Gregory the Great.

Author of numerous monumental works (for example the Door of the Glorification of Mary in the Cathedral of Siena in 1958) , a tireless worker, his work ranges from the monument to the fallen for the city of Busto Arsizio (1951) to that of Pope John XXIII located in Sanctuary of the Madonna del Bosco of Imbersago (1962) . His presence is clearly visible in the private chapel of Pope Paul VI for which he created the altar, the crucifix and also the bronze door. He was defined as the "sculptor of the Popes" because in his long career he had the opportunity to portray all the Popes who succeeded one another on the papal throne from Pius XII (whose bust is preserved in St. Patrick's Cathedral in New York) up to John Paul II.

He shared the high humanity of Pope Roncalli and Pope Montini who posed directly for him and were therefore able to know him personally. The artist's greatest commitment can certainly be attributed to the large doors and portals of Cathedrals and Churches including, in addition to the aforementioned door of the Cathedral of Siena, the bronze door of the Church dedicated to Saint Paul in Damascus, the bronze of the Cathedral of Troia (1971) the door of the Cathedral of Cava dei Tirreni, a large panel for the facade and the three doors of the Cathedral of St. Mary in San Francisco, California, created together with other works exhibited inside the church in the early 1970s, the three doors of the Cathedral of S. Nicolò in Lecco (1975) , a bronze panel depicting the Madonna and Child among the SS. Gioachimo, Anna, Antonio di Padova and Rita da Cascia in the church of San Gioachimo in Milan and, finally, the door of San Paolo outside the walls in Rome, inaugurated in 2000 when the sculptor was already 83 years old. His is also the statue of Maternity located in the Parco della Mamma in Tordibetto di Assisi, commissioned by the parish priest Don Otello Migliosi in 1970 on the occasion of the fourteenth anniversary of the institution of the festival.

He was also the author of a very notable number of commemorative medals and coins which marked the most significant stages of the pontificates from Pius XII to John Paul II. A large number of portraits, Stations of the Cross and bronze plaques can also be attributed to him, mostly of religious subjects; the works created during the papacy of Paul VI are of great interest. A bronze half-bust of the Madonna and Child is preserved in the Mother Church of Tuglie (Lecce) , a gift from the Tugliese painter Cosimo Sponziello (1915-2005) , a friend of the sculptor.
The fisherman's ring chosen by Pope Francis at the beginning of his pontificate is also his work.

He died in 2004; he is buried in the monumental cemetery of Lugo.
